INTRODUCCIÓN: Candida glabrata es una levadura oportunista que causa infecciones fúngicas invasivas en pacientes inmunosuprimidos con una tasa de mortalidad muy alta debido a la resistencia que este microorganismo ha desarrollado a los antifúngicos; esta levadura posee MIC muy alta para fluconazol, y en los últimos años se han aislados cepas resistentes a la caspofungina (equinocandina de elección como tratamiento de primera línea) debido a mutaciones en genes FKS cuya sobreexpresión es mediada por la vía Calcineurina/Calmodulina. Por lo anterior se hace necesario realizar una mutante del factor de transcripción CRZ1 y evidenciar retornar de un fenotipo resistente a sensible.
OBJETIVO: Obtener mutantes de Candida glabrata resistentes a Caspofungina aisladas de muestras clínicas para el factor de transcripción CRZ1 mediante el sistema CRISPR-Cas9.
METODOLOGÍA: se realizó una búsqueda informática de la literatura sobre el sistema CRISPR-Cas en levaduras. Además, se diseñó un plásmido con la secuencia del sgRNA complementario a una fracción de la secuencia del gen CRZ1, y con el sistema CRISPR-Cas9 para transfectar las levaduras y seleccionar las cepas mutadas a través de un marcador de selección fluorescente.
RESULTADOS: En la búsqueda sistemática se analizaron 39 artículos en los cuales, se obtuvo información sobre el sistema CRISPR-Cas en levaduras de importancia clínica e industrial, se documentó las variantes descritas en levaduras para el uso del sistema CRISPR/Cas. La construcción bioinformática de C. glabrata, se obtuvieron dos sgRNA con score de 89 y 71%, un mapa del constructo del plásmido con el sistema CRISPR más el marcador YFP y el SgRNA, la generación del mutante no se logró realizar por incumplimiento de la casa comercial.
CONCLUSIÓN: El sistema CRISPR es un sistema muy versátil y eficientes, que permite realizar la edición del genoma de diferentes organismos celulares. Para lograr el éxito de la edición, es muy importante un buen diseño bioinformática que garantice la especificidad del sistema, y el reconocimiento adecuado de la Cas9 de la secuencia diana que se quiere editar.
Abstrakt
INTRODUCTION: Candida glabrata is an opportunistic yeast that causes invasive fungal infections in immunosuppressed patients with a very high mortality rate due to the resistance that this microorganism has developed to antifungals; This yeast is very useful for the use of the word castrofungin. Therefore, it is necessary to carry out a mutation of the transcription factor CRZ1 and to show a phenomenon resistant to a sensitive one.
OBJECTIVE: To obtain mutants of Candida glabrata in a Caspofungin, in a sample of samples for the transcription factor CRZ1 through the CRISPR-Cas9 system.
METHODOLOGY: a computer search of the literature on the CRISPR-Cas system in yeast was carried out. In addition, a plasmid was designed with the sequence of the complementary sgRNA and a part of the CRZ1 gene sequence, and with the CRISPR-Cas9 system for the treatment of the yeasts and the selection of the mutated strains through a fluorescent selection marker. .
RESULTS: In the systematic search, 39 articles will be analyzed in which information on the CRISPR-Cas system will be obtained in yeasts of clinical and industrial importance, the variants described in yeasts will be documented for the use of the CRISPR / Cas system. The bioinformatic construction of C. glabrata, we obtained two sgRNA with a score of 89 and 71%, a map of the plasmid construct with the CRISPR system plus the YFP marker and the SgRNA, the generation of the mutant can not be performed on the contrary Comercial House.
CONCLUSION: The CRISPR system is a very versatile and efficient system that allows the editing of the genome of different cellular organisms. To achieve the success of the edition, a good bioinformatic design is very important that guarantees the specificity of the system, and the adequate recognition of the Casina of the sequence.
