Evaluación de la actividad antifúngica del péptido LfcinB (21-25)pal RWQWRWQWR derivado de la lactoferricina bovina en biopelículas de Candida auris resistente a fluconazol

Candida auris es un patógeno fúngico oportunista que se ha diseminado globalmente en los últimos años causando infecciones nosocomiales asociadas a altas tasas de mortalidad especialmente en pacientes inmunocomprometidos. Se caracteriza por ser multidrogo resistente a antifúngicos como fluconazol, Anfotericina B o caspofungina por medio de mecanismos en donde se destaca la formación de biopelículas. En este trabajo, se evaluó la actividad antifúngica del péptido LfcinB (21-25)pal RWQWRWQWR derivado de la lactoferricina bovina en biopelículas de Candida auris resistente a fluconazol. Con el fin de establecer las condiciones óptimas de crecimiento de la levadura, se evaluó la formación de la biopelícula frente a diferentes concentraciones de glucosa (0%, 5%, 15%, 25% y 30%), de inóculo (106 ,107 y 108 células/mL) y tiempo de incubación (24 y 48 horas), en placas de 96 pozos a los cuales se les agregó resazurina para la lectura de la fluorescencia obtenida en el equipo FLUOstar® Omega. Una vez establecidos estos parámetros, se procedió a evaluar el péptido de dos formas: (i) agregándolo junto con el inóculo pre incubado por 60 minutos a 37°C, y (ii) agregándolo posterior a la pre incubación del inóculo a las condiciones descritas. Los resultados obtenidos mostraron que a diferentes concentraciones de inóculo y de glucosa no se ve afectada la cantidad de biomasa producida por la biopelícula en C. auris. Además, no se observaron diferencias de crecimiento entre las 24 y 48 horas de incubación. La acción del péptido fue similar para las dos condiciones evaluadas, evidenciándose una reducción en la formación de la biopelícula para ambos casos de aproximadamente 75%. Se concluye que el péptido LfcinB (21-25)pal efectivamente previene la formación de la biopelícula de C. auris en etapas tempranas de crecimiento, lo cual, lo convierte en un candidato promisorio para el tratamiento de infecciones de C. auris asociadas a biopelículas.

Candida auris is an opportunistic fungal pathogen that has spread globally in recent years causing nosocomial infections associated with high mortality rates especially in immunocompromised patients. It is characterized by multidrug resistant to antifungals such as fluconazole, amphotericin B or caspofungin through mechanisms where the formation of biofilms is highlighted. In this work, the antifungal activity of the peptide LfcinB (21-25) pal RWQWRWQWR derived from bovine lactoferricin in biofilms of Candida auris resistant to fluconazole was evaluated. In order to establish the optimal growth conditions of the yeast, the formation of the biofilm was evaluated against different concentrations of glucose (0%, 5%, 15%, 25% and 30%), of inoculum (106, 107 and 108 cells / mL) and incubation time (24 and 48 hours), in 96-well plates, to which resazurin was added for reading the fluorescence obtained in the FLUOstar® Omega equipment. Once these parameters were established, the peptide was evaluated in two ways: (i) adding it together with the pre-incubated inoculum for 60 minutes at 37°C, and (ii) adding it after the pre-incubation of the inoculum to the described conditions . The results obtained showed that the amount of biomass produced by the biofilm in C. auris is not affected at different concentrations of inoculum and glucose. In addition, no growth differences were observed between 24 and 48 hours of incubation. The action of the peptide was similar for the two conditions evaluated, evidencing a reduction in the formation of the biofilm for both cases of approximately 75%. It is concluded that the peptide LfcinB (21-25) pal effectively prevents the formation of the biofilm of C. auris in early stages of growth, which makes it a promising candidate for the treatment of C. auris infections associated with biofilms.
