Correlation of the PCR and ELISA diagnostic tests for the detection of bovine leukosis virus in different animal speciesResumen
El virus de la leucosis bovina (VLB), es el agente etiológico de la leucosis bovina enzootica (LBE), enfermedad caracterizada por ser linfoproliferativa y contagiosa. El VLB infecta de manera natural al ganado bovino, sin embargo, infecta a especies como los búfalos y el ganado ovino. Adicional el VLB se ha encontrado en humanos en sangre periférica, en tejido mamario con y sin cáncer de mama y en pulmón, planteando la propuesta de que este virus pueden ser de carácter zoonótico.
En el transcurso de la investigación con VLB se han encontrado resultados falsos negativos al realizar IGDA y ELISA para el diagnóstico del virus, por lo tanto, se recomienda utilizar la técnica de PCR para contrarrestar la poca sensibilidad de las pruebas serológicas. En la literatura se han reportado algunos estudios que se han realizado para evaluar la concordancia de pruebas diagnósticas para la detección del VLB como PCR y ELISA, dando como resultado que existen diferencias en el diagnóstico según las pruebas utilizadas, y concuerda con que la PCR es la técnica más adecuada para encontrar el VLB o segmentos de él, mientras que las pruebas de ELISA buscan anticuerpos lo cual puede llevar a un mayor número de resultados falsos negativos.
Teniendo en cuenta estos antecedentes el objetivo de este trabajo fue evaluar la concordancia para el diagnóstico del VLB entre las pruebas de ELISA y PCR en diferentes especies. Para esto, se utilizó una ELISA de bloqueo de la casa comercial INGENAZA diseñada para detectar anticuerpos anti-gp51 del VLB a partir de muestras de suero o de leche, y por otro lado, pruebas de PCR dirigidas al segmento génico gag del virus. El trabajo se realizó con muestras obtenidas de cuatro especies (68 muestras de bovinos, 95 de ovinos, 42 de búfalos y 169 de humanos) a las cuales se les realizaron las pruebas previamente mencionadas. Para las serologías se encontraron 25% de muestras positivas para VLB provenientes de bovinos, y ninguna positiva para las muestras provenientes de otras especies; mientras por PCR, se encontró que el 57% de las muestras de bovinos fueron positivas para el virus, 18% de las muestras de ovinos, 21% de búfalos y 54% de humanos.
Abstract
Bovine leukosis virus (VLB) is the etiologic agent of enzootic bovine leukosis (LBE), a disease characterized by being lymphoproliferative and contagious. The VLB naturally infects cattle, however, it infects species such as buffalo and sheep. Additionally, VLB has been found in humans in peripheral blood, in breast tissue with and without breast cancer and in lung, raising the proposal that this virus may be zoonotic in nature.
During the investigation with VLB false negative results have been found when performing IGDA and ELISA for the diagnosis of the virus, therefore, it is recommended to use the PCR technique to counteract the low sensitivity of serological tests. Some studies have been reported in the literature that have been conducted to evaluate the concordance of diagnostic tests for the detection of BVL as PCR and ELISA, resulting in differences in the diagnosis according to the tests used, and agrees that the PCR is the most appropriate technique to find the VLB or segments of it, while ELISA tests look for antibodies which can lead to a greater number of false negative results.
Taking into account these antecedents, the objective of this work was to evaluate the concordance for the diagnosis of VLB between ELISA and PCR tests in different species. For this, a blocking ELISA of the INGENAZA commercial house designed to detect anti-gp51 antibodies from VLB from serum or milk samples was used, and on the other hand, PCR tests directed to the gag gene segment of the virus. The work was carried out with samples obtained from four species (68 samples of cattle, 95 of sheep, 42 of buffalo and 169 of humans) to which the previously mentioned tests were performed. For serologies, 25% of samples positive for VLB from cattle were found, and none positive for samples from other species; while by PCR, it was found that 57% of the bovine samples were positive for the virus, 18% of the sheep samples, 21% of buffalo and 54% of humans
