Effect of the use of white rot fungi Pleurotus ostreatus and Phanerochaete sp. in the biotransformation of oxodegradable straws pretreated with photolysisSommario
El polietileno (PE) es el plástico más común. El polietileno de baja densidad (PEBD) es el más empleado, especialmente para el uso de implementos de un solo uso como pitillos, bolsas, platos, cubiertos, entre otros. Sin embargo, el excesivo uso y mal manejo de estos desechos ha llevado a la afectación de diferentes ecosistemas, en especial el marino, causando daño a los animales y biota en general. Dentro de los problemas más reconocidos están el enredo, estrangulamiento y asfixia.
Hace unas décadas, se buscaron alternativas para el manejo de estos plásticos, como el uso de aditivos prooxidantes, los cuales son complejos de metales de transición y/o estearatos metálicos que aceleran la fotooxidación. A pesar de esto, se ha encontrado que estos aditivos debilitan el material y permiten que se fraccione en microplásticos y nanoplásticos, los cuales son difíciles de identificar a simple vista, y por su tamaño y fragilidad se han distribuido a los demás ecosistemas, reportándose en literatura casos de citotoxicidad por inhalación o consumo.
Por ende, se han buscado otras estrategias que permitan degradar los plásticos, como el uso de luz ultravioleta que debilita el material y puede hacerlo más accesible a colonización por microorganismos gracias a la adición de grupos químicos polares en su superficie. Dentro de los microorganismos que se han encontrado que tienen la capacidad de degradar polímeros, están
los hongos de podredumbre blanca, los cuales poseen enzimas como las MnP y LiP que degradan polímeros vegetales como la lignina, que posee estructuras similares a los polímeros plásticos como los enlaces C-C (carbono-carbono), por ello pretenden ser una alternativa prometedora.
En este trabajo se propuso el uso de luz ultravioleta como pretratamiento y la utilización de hongos de podredumbre blanca Pleurotus ostreatus y Phanerochaete sp., para realizar biotransformación sobre láminas de pitillo de PEBD con medidas de 1 cm x 1 cm, utilizando sustratos como la borra de café y corteza de pino para favorecer el metabolismo de los hongos mencionados. A las láminas de pitillo de PEBD oxodegradables prístino, postfotólisis y postratamiento biológico, se les realizaron determinaciones de los grupos funcionales mediante FTIR-ATR, cambios en la rugosidad y topografía mediante SEM y AFM, así como variación de
hidrofobicidad mediante el ángulo de contacto estático (SCA) y peso. Los resultados mostraron que el pretratamiento con luz ultravioleta favorece el aumento del SCA y rugosidad sobre la superficie de las láminas de pitillo de PEBD oxodegradable, lo que concluye que gracias a este pudo darse una fotoxidación. Luego del tratamiento biológico, los hongos realizaron cambios más notorios en cuanto al aumento de la rugosidad, del SCA y presencia de grupos químicos polares en la mayoría de los tratamientos con Pleurotus ostreatus y Phanerochaete sp, la ausencia de grupos funcionales OH en los tratamientos Po I de 5g, Po I de 15 g, Pc I de 10g, Pc I de 20g y Pc NI de 10 g pudo darse por eliminación de estos, al realizar previo lavado con Tween 80. El
control de Phanerochaete sp. que no tenía sustratos ni láminas de pitillo de PEBD oxodegradable irradiadas con luz ultravioleta Pc NI 0 g, no presentaron señales concretas ni confirmatorias en el espectro realizado con FTIR-ATR, por lo que se pudo evidenciar que la biotransformación de este hongo hacia las láminas de pitillo de PEBD oxodegradables depende de la presencia de
sustrato y del pretratamiento que se les realice a estas. En este estudio, no se observaron resultados importantes en cuanto a la variación de peso en la mayoría de las láminas de pitillo de PEBD oxodegradables postfotólisis ni luego del tratamiento biológico, pues se evidenciaron únicamente en los tratamientos de Phanerochaete sp. Pc I de 10 g, Pc I de 15 g y Pc I de 20 g, lo cual
pudo darse en función a diferencias estructurales del material por el tiempo que llevan desde su fabricación ya que pueden variar las propiedades mecánicas del plástico en comparación con las demás láminas de PEBD utilizadas. Otra razón es porque efectivamente la actividad enzimática del hongo realizó cambios al reorganizar molecularmente las láminas de pitillo de PEBD
oxodegradable evidenciando cambios de masa. Los tratamientos que mostraron variación significativa de todos los parámetros evaluados fueron los del tratamiento de Phanerochaete sp. de P.c I de 10g y 15g mostrando cambios químicos, lo que puede considerarse como biotransformación.
Astratto
Polyethylene (PE) is the most common plastic. Low-density polyethylene (LDPE) is the most widely used, especially for the use of single-use implements such as straws, bags, plates, cutlery, among others. However, the overuse and mismanagement of this waste has affected different ecosystems, particularly marine ecosystems, causing damage to animals and biota in general. Among the most recognized problems are entanglement, strangulation and suffocation. Some decades ago alternatives were sought for the handling of these plastics, such as the use of prooxidant additives, which are complexes of transition metals and / or metal stearates that accelerate photooxidation. Despite this, it has been proven that these additives weaken the material and allow it to be divided into microplastics and nanoplastics, which are difficult to identify with the naked eye, and due to their size and fragility they have been distributed to other ecosystems, reports in bibliographic cases of cytotoxicity by inhalation or consumption.
Therefore, other strategies have been sought that allow the degradation of plastics, such as the use of ultraviolet light that weakens the material and may make it more accessible to colonization by microorganisms thanks to the addition of polar chemical groups on their surface. The microorganisms that have been found to have the ability to degrade polymers, there are white rot fungi, which have enzymes such as MnP and LiP that degrade plant polymers such as lignin, which has structures similar to plastic polymers such as C-C (carbon-carbon) bonds, therefore claim to be a promising alternative. In this study, the use of ultraviolet light as a pretreatment and the use of white rot fungi Pleurotus ostreatus and Phanerochaete sp., werev carry out to do biotransformation on sheets of LDPE scab with measurements of 1 cm x 1 cm, using substrates such as coffee erase and pine bark to promote the metabolism of the fungi mentioned. The sheets of pristine oxodegradable LDPE scrap, post-photolysis and biological post-treatment were determined by FTIR-ATR, changes in roughness and topography by SEM and AFM, as well as changes in hydrophobicity by static contact angle (SCA) and weight. The results showed that pre-treatment with ultraviolet light favors the increase of SCA and roughness on the surface of oxodegradable LDPE scraps, which concluded that photooxidation was possible. After the biological treatment, the fungi made more noticeable changes in terms of increased roughness, SCA and presence of polar chemical groups in most treatments with Pleurotus ostreatus and Phanerochaete sp, the absence of functional groups OH in the treatments Po I of 5g, Po I of 15g, Pc I of 10g, Pc I of 20g and Pc NI of 10 g could be given by elimination of these, when performed prior washing with Tween 80. The control of Phanerochaete sp. which did not have substrates or oxodegradable LDPE scraps irradiated with ultraviolet light Pc NI 0 g, did not show concrete or confirmatory signals in the spectrum performed with FTIR-ATR, so it was possible to demonstrate that the biotransformation of this fungus towards the LDPE scraps oxodegradable depends on the presence of substrate and the pretreatment that is done to them. In the present study, no significant results were observed regarding weight change in most of the oxodegradable LDPE scraps after photolysis or after biological treatment, as they were only evident in the treatments of Phanerochaete sp. Pc I of 10 g, Pc I of 15 g and Pc I of 20 g, which could be due to structural differences of the material due to the time taken since manufacture, as the mechanical properties of the plastic may vary compared to the other LDPE sheets used. Another reason is that indeed the enzymatic activity of the fungus made changes by molecularly rearranging the sheets of LDPE scraps oxodegradable evidencing changes in mass. The treatments that showed significant variation in all evaluated parameters were those of Phanerochaete sp. P. c I of 10g and 15g showing chemical changes, which can be considered biotransformation.
