En los últimos años, el estudio de probióticos para el tratamiento de la Enfermedad
Diarreica Aguda (EDA) en niños ha sido de gran interés debido a múltiples estudios que
sugieren que estos poseen actividad anti-rotavirus y poseen la capacidad de reducir el
tiempo de la diarrea. El grupo de investigación en virología de la Pontificia Universidad
Javeriana ha realizado estudios de la actividad anti-rotavirus y del efecto citotóxico en
células intestinales C2BBe1 ejercidos por proteínas PEGiladas procedentes del secretoma
de Bifidobacterium adolescentis a partir de los cuales se obtuvo una reducción de la
infección viral, no se observó efecto citotóxico y particularmente, se observó que podrían
producir un aumento de la proteína de unión celular ocludina. Así, a partir de lo anterior
surgió la pregunta de si estas proteínas pueden internalizarse en las células para producir
el efecto de aumento de ocludina, al igual que la necesidad de cuantificar este efecto,
recordando además, que están recubiertas de polietilenglicol de alta densidad y por tanto,
teóricamente no deberían entrar. A partir de lo anterior, en este trabajo se buscó evaluar
la capacidad de internalización de las proteínas PEGiladas del secretoma de B.
adolescentis (PPEGBa) en las células intestinales polarizadas C2BBe1, por medio del
marcaje fluorescente y observación de las proteínas, además de su efecto en la expresión
de la proteína ocludina por medio de la cuantificación de ocludina total por fluorometría.
Se determinó que las PPEGBa se acumulan en la periferia celular y además promueven
la expresión de la proteína de unión celular ocludina. Estos resultados sugieren que las
PPEGBa podrían tener la capacidad de fortalecer la barrera intestinal.
Astratto
In recent years, the study of probiotics as treatment for Acute Diarrhoeal Disease (ADD) in kids has been of great interest due to multiple studies that suggest that they possess anti-rotavirus activity and the ability to reduce the time of diarrhea. The virology research group of the Pontificial Xaverian University has carried out studies of the anti-rotavirus activity and the cytotoxic effect in intestinal C2BBe1 cells exerted by PEGylated proteins from the secretome of Bifidobacterium adolescentis, from which was observed a reduction in the viral infection and no cytotoxic effects, it was also particulary observed that they could produce an increase in tight juction protein occludin. Thus, taking in consideration these previous results, the question whether these proteins can be internalized into cells to produce to produce the effect of increasing occludin arose, as well as the need to quantify this effect, remembering that they are coated with high-density polyethylene glycol and therefore, theoretically they should not enter. Based on the above, this work sought to evaluate the internalization capacity of the PEGylated proteins from the secretome of B. adolescentis (PPEGBa) in C2BBe1 polarized intestinal cells, by means of the fluorescent labelling and observation of proteins, in addition to their effect in the expression of occludin by means of the quantification of total occludin by fluorometry. It was determined that PPEGBa accumulate in the cell periphery and also promote the expression of the tight junction protein occludin. These results suggest that PPEGBa may have the ability to strengthen the intestinal barrier.
