Evaluation of the stability of a biotechnological interest extract from the thermoacidophilic microalgae Galdieria sp. USBA -GBX 832Resumen
Los microorganismos extremófilos son de alto interés en biotecnología ya que producen metabolitos que soportan diferentes condiciones como altos y bajas temperaturas, variaciones en el pH, altas presiones, otorgándoles estabilidad y resistencia a procesos realizados en la industria. Por otro lado, las condiciones en las que crecen disminuyen las probabilidades de contaminación. Estos metabolitos podrían ser usado en diferentes industrias como la energética, de alimentos, farmacéutica y cosmecéutica, gracias a las propiedades biológicas que presentan. En particular la microalga termoacidófila Galdieria sp. USBA -GBX 832, produce metabolitos como carotenoides, esteroles, ficocianinas, ácidos grados poliinsaturados, fosfolípidos, entre otros, los cuales pueden presentar actividades antioxidantes, antiinflamatorias, antienvejecimiento y protección a rayos UV, indicando el potencial que tiene este microorganismo en la industria cosmecéutica. Por lo tanto, para comprobar la estabilidad un extracto obtenido a partir de la biomasa de Galdieria sp. USBA -GBX 832, se realizó la evaluación de la estabilidad preliminar, natural y acelerada de tres lotes de producción bajo temperaturas de 4, 25, 40, 45°C y ciclos de temperaturas entre 4 y 40°C durante 3 meses. Se realizaron muestreos periódicos de los lotes y se documentó los cambios en el color, olor, aspecto, sensación al tacto, así como pruebas microbiológicas para el recuento de microorganismos mesófilos, pruebas de presencia o ausencia de microorganismos patógenos y, finalmente se midió las variaciones en la actividad antioxidante usando la técnica del radical ABTS. Los resultados evidenciaron que los tres lotes de extractos de la microalga no presentaron cambios en sus características organolépticas a excepción del olor, el cual se intensificó a través del tiempo. Por otro lado, las pruebas microbiológicas determinaron ausencia de mesófilos, hongos, levaduras, y patógenos en todos los extractos. Finalmente, en cuanto a la actividad antioxidante, esta disminuyó en la semana 1 en todos los lotes, manteniéndose estable durante el resto del tiempo evaluado. Concluyendo que los extractos de sp. USBA -GBX 832 son estables en sus propiedades organolépticas, microbiológicas y de actividad biológica.
Abstract
Extremophilic microorganisms are of high interest in biotechnology since they produce metabolites that withstand different conditions such as high and low temperatures, variations in pH, high pressures, giving them stability and resistance to processes carried out in industry. On the other hand, the conditions in which they grow reduce the chances of contamination. These metabolites could be used in different industries such as energy, food, pharmaceutical and cosmeceutical, thanks to the biological properties they present. In particular the thermoacidophilic microalgae Galdieria sp. USBA -GBX 832, produces metabolites such as carotenoids, sterols, phycocyanins, polyunsaturated fatty acids, phospholipids, among others, which can present antioxidant, anti-inflammatory, anti-aging and UV protection activities, indicating the potential that this microorganism has in the cosmeceutical industry. . Therefore, to check the stability, an extract obtained from the biomass of Galdieria sp. USBA -GBX 832, the evaluation of the preliminary, natural and accelerated stability of three production batches was carried out under temperatures of 4, 25, 40, 45 ° C and temperature cycles between 4 and 40 ° C for 3 months. Periodic samplings of the batches were carried out and the changes in color, odor, appearance, and sensation to the touch were documented, as well as microbiological tests for the count of mesophilic microorganisms, tests of the presence or absence of pathogenic microorganisms and, finally, the variations were measured. in antioxidant activity using the ABTS radical technique. The results showed that the three batches of microalgae extracts did not show changes in their organoleptic characteristics except for the smell, which intensified over time. On the other hand, the microbiological tests determined the absence of mesophiles, fungi, yeasts, and pathogens in all the extracts. Finally, in terms of antioxidant activity, it decreased in week 1 in all batches, remaining stable during the rest of the time evaluated. Concluding that the extracts of sp. USBA -GBX 832 are stable in their organoleptic, microbiological and biological activity properties.
