Evaluación de bacterias sulfato reductoras presentes en reactores pasivos durante la remediación de drenajes ácidos de minas

Resumen
Los biorreactores pasivos son utilizados para la remediación de drenajes ácidos de minas (DAM). sin embargo, se desconoce como el tiempo de retención hidráulica (TRH). el espacio y el tiempo de operación de los reactores pueden afectar la dinámica y la actividad de las bacterias sulfato reductoras (BSR). Para ello se diseñaron 7 biorreactores pasivos de flujo ascendente con 3 TRH diferentes (1. 2 y 4 días). En el efluente de los reactores se monitoreo: el pH. potencial de óxido reducción (ORP). concentración de sulfato, sulfuro, iones metálicos disueltos, y adicionalmente, se cuantificaron las BSR mediante el número más probable (NMP) de BSR y la amplificación del gen dsr.A por qPCR. Para cuantificar las BSR en la mezcla reactiva (mediante el XMP y qPCR) y determinar la actividad microbiana general (enzimas deshidrogenasas y celulolíticas). se realizó el sacrificio de las columnas (semanas 8. 17 y 36) durante los cuales se muestreó la mezcla reactiva en núcleos de aproximadamente 20 cm según la altura del reactor (sección baja, media y alta). Los recuentos de BSR en el efluente estuvieron afectados significativamente por el TRH y el tiempo de operación. Por otro lado, el recuento de BSR en la mezcla reactiva no estuvo influenciado significativamente por el TRH. pero si por las alturas del reactor y el tiempo de operación. La actividad de las BSR fue evaluada en términos de la tasa de remoción de sulfato, producción de sulfuro y porcentaje de remoción de metales. El TRH tuvo efecto sobre la actividad de las BSR en términos de tasa de remoción de sulfato y producción de sulfuro: tiempos coitos permiten mayores tasas de remoción de sulfato y menor producción de sulfuro. La actividad de las BSR en términos del porcentaje de remoción de metales no mostró diferencias para Zn+2 y Fe'2 en los TRH de 2 y 4 días. Sin embargo, no se puede considerar un indicador absoluto dado que la remoción de metales también se puede dar por la formación de oxi-hidróxidos y carbonatos. El TRH. el tiempo y la posición en el reactor tuvieron efecto significativo sobre la actividad deshidrogenasa. Mientras que en el caso de la actividad celulolítica solo tuvo efecto el tiempo de operación.
Abstract
Passive bioreactors are used for the remediation of acid mine drainage (AMD), however, it is unknown how the hydraulic retention time (HRT). The space and time of operation of the reactors may affect the dynamics and activity sulphate-reducing bacteria (SRB). For this purpose. 7 up flow passive bioreactors were designed with 3 different HRT (1. 2 and 4 days), hi the effluent were measured: pH. Redox potential (ORP). Sulfate, dissolved metal ions (Fe~2. Zn+2 and Mn;2) and sulfides concentration: also the SRB were quantified through two approaches: the most probable number (MPN) of SRB and the amplification of the dsrA gene by qPCR. With the aim of quantify the SRB in the reactive mixture (using the MPN and qPC'R) and determine the general microbial activity (dehydrogenases and cellulolytic enzymes), bioreactors were sacrificed (weeks 8. 17 and 36) and the reactive mixture were sampled into cores of approximately 20 cm depending on the height of the reactor (low. medium and high section) The number of SRB in the effluent was significantly affected by HRT and the operation time. Furthermore, the number of SRB in the reactive mixture was not significantly influenced by the HRT. The position in the reactor and the operation time influenced significantly on the number of SRB in the reactors. The SRB activity was evaluated in terms of the sulfate removal rate, sulfide production and metal removal. The HRT had a significantly effect on the activity of the SRB in terms of sulfate removal rate and sulfide production: short times allow higher removal rates and lower production sulfate sulfur. The activity of the SRB showed no differences between Fe+2 and Zn+2 removal for 2 and 4 d of HRT. However, metal removal cannot be considered an absolute indicator since that procedure can also be carried out by the formation of oxy-hydroxides and carbonates. The HRT. Time of operation and the heights in the reactor had significant effect on dehydrogenase activity. While in the case of cellulolytic activity only the operation time have a significant effect.
