El uso de nemátodos entomopatógenos (NEPs) como controladores biológicos en cultivos agrícolas es una alternativa al uso de pesticidas de síntesis química, los cuales han demostrado ser nocivos para el medio ambiente y el ser humano y además, han perdido efectividad por el fenómeno de resistencia. Para suplir la demanda de los NEPs en campo se ha desarrollado el método de producción in vitro, el cual a diferencia del convencional (in vivo) tiene mayor rendimiento y es menos dispendioso. Dicho método requiere el diseño de un medio de cultivo con composición similar al interior de las larvas de insectos y la capacidad de la bacteria simbionte (para Heterorhabditis sp. es Photorhabdus luminescens) de digerir la biomasa del insecto, en este caso los componentes del medio diseñado. Una de las mayores dificultades del método es determinar el tiempo de inoculación de juveniles infectivos (JIs) debido el cambio de metabolismo al cultivar la bacteria in vitro, donde la bacteria pasa de ser apta para digerir el medio (fase I) a no ser apta para esta función (fase intermedia y II). Esta capacidad está dada principalmente por la producción de enzimas líticas, antibióticos y “señales de alimentación”. Con base en lo anterior, en este trabajo se evaluó la cinética de crecimiento, patogenicidad en Galleria mellonella (Lepidoptera: Pyralidae) y cambio de fase I a intermedia y II de Photorhabdus luminescens akhurstii SL0708 en medio de producción in vitro de Heterorhabditis indica SL0708; con el fin de determinar en qué momento es oportuno inocular los JIs. Para lograr lo anterior se realizó una curva de crecimiento microbiano en medio de producción in vitro de H. indica SL0708 durante 96h a 28ºC y 150 rpm. Mediante un muestreo destructivo de cada 12 horas, se determinó la formación de biomasa (UFC/ml), características de las colonias en medio NBTA, luminiscencia cualitativamente y cuantitativamente (UAL: unidades arbitrarias de luminiscencia), pH y azúcares reductores equivalentes a glucosa (g/L). Adicionalmente, se hizo una valoración de asimilación de carbohidratos utilizando API 20NE y de la patogenicidad en larvas del último instar de G. mellonella. Cada ensayo se realizó por triplicado y la curva se hizo 3 veces en el tiempo. Como resultado se obtuvo que la cinética de P. luminescens akhurstii SL0708 en el medio evaluado consiste en una fase exponencial de 84 horas con dos velocidades de crecimiento diferentes y un periodo diaúxico entre estas, y una fase estacionaria a partir de las 84 horas. De acuerdo con la literatura revisada no se han reportado estudios con resultados cinéticos similares. Por otro lado se determinó que la diferencia de las velocidades de crecimiento coincide con el cambio de fase I a intermedia y II, y con el consumo de la mayoría de la glucosa (77,4%). Lo anterior resalta la importancia de la glucosa como sustrato que mantiene la fase I, la cual tiene una velocidad de crecimiento menor a la de la fase intermedia y II. Por último se observó que el cambio de fase I a intermedia y II de la bacteria, no tiene efecto sobre la patogenicidad en larvas de G. mellonella y la asimilación de carbohidratos.
Abstract
The use of entomopathogenic nematodes (EPNs) as biological controllers in agricultural crops is an alternative to the use of pesticides of chemical synthesis, which have shown to be harmful to the environment and the human being, and also have lost effectiveness due to the phenomenon of resistance. To meet the demand for EPNs in the field, the in vitro production method has been developed, which, unlike the conventional one (in vivo), has a higher yield and is less expensive. This method requires the design of a culture medium with composition similar to the interior of the insect larvae and the ability of the symbiont bacterium (for Heterorhabditis sp., Photorhabdus luminescens) to digest the biomass of the insect, in this case the components of the medium designed. One of the major difficulties of the method is to determine the inoculation time of infective juveniles (IJs) due to the change of metabolism when growing the bacterium in vitro, where the bacterium goes from being able to digest the medium (phase I) to not being suitable for this function (intermediate phase and II). This capacity is mainly given by the production of lytic enzymes, antibiotics and "feeding signals". Based on the foregoing, in this work the kinetics of growth, pathogenicity in Galleria mellonella (Lepidoptera: Pyralidae) and change from phase I to intermediate and II of Photorhabdus luminescens akhurstii SL0708 in in vitro production medium of Heterorhabditis indica SL0708 were evaluated; in order to determine when it is appropriate to inoculate the IJs. To achieve the above, a microbial growth curve was performed in the in vitro production medium of H. indica SL0708 for 96h at 28ºC and 150 rpm. By means of a destructive sampling every 12 hours, the formation of biomass (CFU/ml), characteristics of the colonies in NBTA medium, luminescence qualitatively and quantitatively (ALU: arbitrary units of luminescence), pH and reducing sugars equivalent to glucose were determined (g/L). Additionally, an assessment of the assimilation of carbohydrates was made using API 20NE and of the pathogenicity in larvae of the last instar of G. mellonella. Each test was performed in triplicate and the curve was made 3 times in time. As a result it was obtained that the kinetics of P. luminescens akhurstii SL0708 in the evaluated medium consists of an exponential phase of 84 hours with two different growth rates and a diauxic period between these, and a stationary phase after 84 hours. According to the reviewed literature, no studies with similar kinetic results have been reported. On the other hand, it was determined that the difference in growth rates coincides with the change from phase I to intermediate and II, and with the consumption of the majority of glucose (77.4%). This highlights the importance of glucose as a substrate that maintains phase I, which has a growth rate lower than that of the intermediate phase and II. Finally, it was observed that the change from phase I to intermediate and II of the bacterium, has no effect on the pathogenicity of G. mellonella larvae and the assimilation of carbohydrates.
