Evaluación de la expresión de la proteína recombinante fenilalanina hidroxilasa humana producida por Lactobacillus plantarum

Resumen
La fenilcetonuria es la amioacidopatía más frecuente dentro de los errores innatos de metabolismo y consiste en la acumulación del aminoácido L-fenilalanina (Phe) en tejidos y plasma a causa de la pérdida de actividad de la enzima fenilalanina hidroxilasa la cual oxida la Phe a L-tirosina. La acumulación de este compuesto tiene un efecto neurotóxico y puede llegar a deteriorar la salud de quien padece la enfermedad si no es tratada a tiempo. Este trabajo de grado tuvo como objetivo evaluar a Lactobacillis plantarum como sistema de expresión de la enzima fenilalanina hidroxilasa humana, con el fin de determinar si la proteína heteróloga puede ser empleada como agente terapéutico contra la enfermedad.
Dentro de los hallazgos obtenidos en este estudio, se obtuvo que Lactobacillus plantarum es sistema de expresión que es fácilmente transformable. Sin embargo, se observaron problemas con el seguimiento de la proteína recombinante mediante el uso del reportero, proteína verde fluorescente. Por otra parte, mediante técnicas moleculares como SDS PAGE y western blot fue posible identificar la presencia de esta enzima, donde los resultados sugieren que la proteína reportera está ausente. Finalmente, midiendo la actividad enzimática del clon positivo para la enzima se pudo concluir que este clon sintetiza una proteína funcional.
Estos resultados muestran la importancia de seguir estudiando el uso de L. plantarum como sistema de expresión. Sin embargo, futuros trabajos se deben enfocar en la confirmación de la producción de la proteína, así como en nuevos reporteros que permitan seguir el proceso de producción de la proteína de una manera más eficiente.
Abstract
Phenylketonuria is the most common aminoacidopathy in innate metabolism errors and consists of the accumulation of the amino acid L-phenylalanine (Phe) in tissues and plasma due to the loss of activity of the enzyme phenylalanine hydroxylase which oxidizes the Phe to L-tyrosine. The accumulation of this compound has a neurotoxic effect and can damage the health of the sufferer if not treated in time. This study aimed to evaluate Lactobacillis plantarum as an expression system of the human enzyme phenylalanine hydroxylase, in order to determine whether heterologous protein can be used as a therapeutic agent against the disease.
Among the findings obtained in this study, it was obtained that Lactobacillus plantarum is system of expression that is easily transformable. However, problems were observed with the tracking of recombinant protein through the use of the reporter, green fluorescent protein. On the other hand, using molecular techniques such as SDS-PAGE and western blot it was possible to identify the presence of this enzyme, where the results suggest that the reporter protein is absent. Finally, by measuring the enzymatic activity of the positive clone for the enzyme, it was concluded that this clone synthesizes a functional protein.
These results show the importance of further studying the use of L. plantarum as an expression system. However, future work should focus on confirming the production of the protein, as well as on new reporters that allow them to follow the process of producing the protein in a more efficient way
