Evaluación de la actividad antifúngica de extractos del género Piper contra Moniliophthora perniciosa, agente causal de Escoba de Bruja en cacao

Colombia ocupa el puesto 10 como uno de los mayores productores de cacao a nivel mundial, que junto con otros cultivos tropicales es de gran importancia en el sector agrícola y económico por sus numerosas características organolépticas y medicinales; sin embargo, estos cultivos se han afectado en razón a las malas prácticas agrícolas, lo cual ha contribuido a la aparición y expansión de enfermedades producidas por hongos, dentro de las cuales la Escoba de Bruja, generada por Moniliophtora perniciosa, ha sido una de las más incidentes en el país. El objetivo de este trabajo fue ofrecer una alternativa a esta problemática, contribuyendo en la búsqueda de agentes fitosanitarios para el control de M. perniciosa a partir de extractos obtenidos de especies del género Piper, considerado un género potencial con actividad antifúngica. Para ello, se realizó es aislamiento y purificación en varios medios de cultivo (PDA, PDA ½, extracto de malta, avena, V8 y Zanahoria) de morfotipos fúngicos asociados a la enfermedad a partir de muestras vegetales con síntomas colectadas en el municipio de Otanche, Boyacá. También se buscó determinar la temperatura y medio cultivo que estimularan el crecimiento y conidiogénesis del posible aislamiento de M. perniciosa y un aislamiento de M. roreri evaluando el desarrollo de las colonias durante 34 días. A pesar de no lograr la producción de conidios se optó por realizar el ensayo de inhibición de crecimiento micelial por el método de dilución en agar en cajas de 12 pozos con M. roreri, esto se hizo evaluando 11 especies de Piper dentro de las cuales se destacaron P. asperiusculum, P. grande, P. statarium, P. artanthe y P. nigrum por inhibir el crecimiento del patógeno en porcentajes superiores al 50%, con lo cual se demostró el gran potencial y las propiedades biológicas de estas especies como sustancias antifúngicas.
Abstract
Colombia ranks 10th as one of the largest producers of cocoa worldwide, which along with other tropical crops is of great importance in the agricultural and economic sector for its numerous organoleptic and medicinal characteristics; However, these crops have been affected due to poor agricultural practices, which has contributed to the appearance and expansion of diseases caused by fungi, among which the Witch Broom, generated by Moniliophtora perniciosa, has been one of the more incidents in the country. The objective of this work was to offer an alternative to this problem, contributing in the search of phytosanitary agents for the control of M. perniciosa from extracts obtained from species of the Piper genus, considered a potential genus with antifungal activity. To this end, isolation and purification was carried out in several culture media (PDA, PDA ½, Malt Extract, oats, V8 and Carrot) of fungal morphotypes associated with the disease from plant samples with symptoms collected in the municipality of Otanche , Boyacá. We also sought to determine the temperature and culture medium that stimulated the growth and conidiogenesis of the possible isolation of M. perniciosa and an isolation of M. roreri evaluating the development of the colonies during 34 days. In spite of not achieving conidium production, we chose to perform the mycelial growth inhibition test by agar dilution method in 12-well boxes with M. roreri, this was done by evaluating 11 Piper species within which They highlighted P. asperiusculum, P. grande, P. statarium, P. artanthe and P. nigrum for inhibiting the growth of the pathogen in percentages higher than 50%, which showed the great potential and biological properties of these species as substances antifungal.
