El aumento de la demanda energética mundial y la utilización de productos petroquímicos en la vida cotidiana, han incrementado las actividades intensivas de exploración, producción y refinamiento de petróleo. Como subproducto de estas actividades se generan gran cantidad de residuos conocidos como lodos aceitosos ó borras aceitosas. Estos residuos representan un peligro para la salud humana y el medio ambiente debido a la presencia de compuestos que pueden ser tóxicos, mutagénicos ó carcinogénicos. En Colombia los lodos aceitosos representan la mayor cantidad de residuos peligrosos producidos. La mayoría de técnicas fisicoquímicas convencionales para el tratamiento de estos residuos pueden ser muy costosas, complejas de implementar y producen altas cargas contaminantes en el ambiente.
La biorremediación que se implementa mediante estrategias de bioestimulación y bioaumentación se ha convertido en una técnica viable para el tratamiento de lodos y borras aceitosas. El objetivo del presente estudio fue evaluar el efecto de la bioaumentación con BioTigerTM, en la degradación de borras aceitosas provenientes de residuos de la industria petrolera, a través de aproximaciones en campo y en laboratorio. En el laboratorio se evaluaron las mismas condiciones de BioTigerTM evaluado en la fase de campo, así como la adición de una inoculación de BioTigerTM y nutrientes a los 30 días de tratamiento. Los resultados obtenidos en laboratorio se compararon con el obtenido en campo. Para esto se evaluó la concentración de hidrocarburos totales de petróleo (TPH) y el número más probable de microorganismos degradadores de hexadecano, empleando el número más probable. La evaluación se realizó en campo durante 204 días y en laboratorio durante 90 días. Así mismo se planteó realizar la confirmación de la identificación taxonómica de las cepas del consorcio de BioTigerTM mediante secuenciación del gen ribosomal 16S.
Durante los estudios se observó la degradación de TPH bajo condiciones de campo y laboratorio. Sin embargo, no se encontraron diferencias significativas en la degradación entre campo y laboratorio ni entre los tratamientos evaluados en laboratorio a los 90 días. Se observó disminución de los recuentos de degradadores en campo y laboratorio hacia los 60 días y una relación entre la disminución de los recuentos y la disminución en el pH. Las amplificaciones del gen ribosomal 16s serán enviadas a Macrogen para su secuenciación
Abstract
The increase of global energy demand and the use of petrochemical products in everyday life increased the intensive activities of oil exploration, production and refining. As a byproduct of these activities, a large amount of a waste known as “oily sludge” or “heavy oily sludge” are generated. These residues represent a danger to human health and the environment due to the presence of compounds that can be toxic, mutagenic or carcinogenic. In Colombia, oily sludge represents the largest amount of hazardous waste produced. Most conventional physicochemical techniques for the treatment of these wastes can be very expensive, complex to implement and produce high pollution loads in the environment.
Bioremediation, implemented through biostimulation and bioaugmentation strategies, has become a viable technique for the treatment of oily sludge and heavy oily sludge. The objective of the present study is to evaluate the effect of bioaugmentation with BioTigerTM on the degradation of heavy oily sludge from residues of the oil industry through an approach on the field and in the laboratory. In the laboratory, the same conditions of BioTigerTM evaluated in the field phase were evaluated, as well as the addition of a BioTigerTM and nutrient inoculation after 30 days of treatment. The results obtained in the laboratory were compared with those obtained on the field. For this, the concentration of total petroleum hydrocarbons (TPH) and the most probable number of hexadecane degrading microorganisms were evaluated, using the most probable number. The evaluation was carried out in the field for 204 days and in the laboratory for 90 days. It was also proposed to confirm the taxonomic identification of the BioTigerTM consortium strains by sequencing the 16S ribosomal gene.
During the studies, the degradation of TPH was observed in field as well in laboratory conditions. However, no significant differences were found in the degradation between field and laboratory or between the treatments evaluated in the laboratory at 90 days. There was a decrease in degrading microorganisms on the field and in the laboratory around 60 days and a relationship between the decrease in in degrader microorganisms and the decrease in pH was observed. Amplifications of the 16s ribosomal gene will be sent to Macrogen for sequencing.
