Production and characterization of the recombinant protein β-fructosidase from Aspergillus oryzae N74 in Pichia pastoris GS115résumé
Los fructooligosacáridos (FOS) son hidratos de carbono de carácter prebiótico que tienen la capacidad de ser asimilados y degradaos únicamente por bacterias benéficas en el tracto gastrointestinal de humanos y animales. Estos polímeros son sintetizados gracias a la unión de moléculas de fructosa a moléculas de sacarosa mediante enlaces (1-2) mediante la acción de enzimas como fructosiltransferasa (FTasa) y β-fructosidasa las cuales son producidas naturalmente por microrganismos como Aspergillus oryzae, Aspergillus japonicas, Aspergillus sydowi, Schawanniomyces occidentalis y Candida utilis. Estas enzimas tienen la capacidad de hidrolizar una molécula de sacarosa y por ende liberar unidades de fructosa y glucosa, posteriormente transfiere las unidades libres de fructosa a aceptores como otra molécula de sacarosa u otro FOS. Sin embargo, la obtención de esta enzima a nivel industrial no es fácil por la complejidad que representa el uso de microrganismos filamentosos. Adicionalmente, estudios previos han demostrado que la enzima FTasa recombinante no ha obtenido buenos rendimientos en la producción de FOS. Por lo tanto, en este trabajo se sintetizó de manera recombinante la proteína β-fructosidasa en P. pastoris GS115 y se evaluó la producción de esta enzima a escala de 10 mL, 100 mL y 1.7 L (biorreactor). Se determinó que el clon Mut+(S23) presenta poca actividad hidrolítica y presenta preferencia hacia la actividad transfructosilante, por ende, esta muestra se seleccionó para ser producida a escala de biorreactor en donde obtuvo el mismo comportamiento pero con leves incrementos en las actividades enzimáticas. Sin embargo, cuando se comparan estos resultados con los reportados en la literatura, los obtenidos experimentalmente son bajos, lo cual se podría explicar a la dificultad que tienen los sustratos de ingresar al sitio activo de la proteína lo cual se evidenció en determinaciones bioinformáticas y al uso de condiciones de reacción no óptimas.
Abstrait
Fructooligosaccharides (FOS) are carbohydrates of a prebiotic nature that can be assimilated and degraded only by beneficial bacteria in the gastrointestinal tract of humans and animals. These polymers are synthesized thanks to the union of fructose molecules to sucrose molecules through "beta" (1-2) bonds through the action of enzymes such as fructosyltransferase (FTase) and β-fructosidase which are naturally produced by microorganisms such as Aspergillus oryzae, Aspergillus japonicas, Aspergillus sydowi, Schawanniomyces occidentalis and Candida utilis. These enzymes can hydrolyze a sucrose molecule and thus release fructose and glucose units, subsequently transfer the free fructose units to acceptors such as another sucrose molecule or another FOS. However, obtaining this enzyme at the industrial level is not easy because of the complexity of the use of filamentous microorganisms. Additionally, previous studies have shown that the recombinant FTase enzyme has not obtained good yields in the production of FOS. Therefore, in this work, the β-fructosidase protein was recombinantly synthesized in P. pastoris GS115 and the production of this enzyme was evaluated on a scale of 10 mL, 100 mL and 1.7 L (bioreactor). It was determined that the Mut + clone (S23) has little hydrolytic activity and has a preference for transfructosilant activity, therefore, this sample was selected to be produced at the bioreactor scale where it obtained the same behavior but with slight increases in enzymatic activities. However, when these results are compared with those reported in the literature, those obtained experimentally are low, which could be explained by the difficulty of the substrates to enter the active site of the protein which was evidenced in bioinformatic determinations and when the use of non-optimal reaction conditions.
