La obesidad es una enfermedad multifactorial, altamente compleja y su prevalencia ha venido en aumento en los últimos años. Una manera de contribuir al conocimiento en esta área es por medio de la evaluación de la expresión génica, y las técnicas de Reacción en Cadena de la Polimerasa (PCR), PCR con Transcriptasa Inversa (RT-PCR) y PCR en tiempo real (qPCR), son los métodos más confiables y utilizados. A pesar de que existen múltiples reportes de genes housekeeping para la línea 3T3-L1, aún se encuentran deficiencias en los diseños de los primers, y esto puede generar resultados poco confiables. Es así como, en este estudio se evaluó la eficacia de una pareja de primers (uno reportado en la literatura y otro diseñado por el autor) para Beta Actina (ACTB), Gliceraldehído 3-fosfato deshidrogenasa (GAPDH) y ARN ribosomal 18s (RNA 18s) en la línea 3T3-L1, con el fin de estandarizar su uso para posteriores montajes experimentales en estas células. Para futuros ensayos se recomienda usar una concentración de 1500ng de ADNc para ACTB-1, ACTB-2, GAPDH-1 y 1000 ng de ADNc RNA18s-1 para obtener resultados confiables y verificables.
Abstract
Obesity is a highly complex multifactorial disease and its prevalence has been increasing in recent years. One way to contribute to knowledge in this area is through the evaluation of gene expression, and the techniques of Polymerase Chain Reaction (PCR), Reverse Transcriptase PCR (RT-PCR) and real-time PCR (qPCR), are the most reliable and widely used methods. Although there are multiple reports of housekeeping genes for the 3T3-L1 line, there are still deficiencies in the primer designs, and this can generate unreliable results. Thus, in this study, the efficacy of a pair of primers was evaluated (one reported in the literature and another designed by the author) for Beta Actin (ACTB), Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and ribosomal RNA 18s (RNA 18s) in the 3T3-L1 line, in order to standardize its use for subsequent experimental setups in these cells. For future assays it is recommended to use a concentration of 1500ng of cDNA for ACTB-1, ACTB-2, GAPDH-1 and 1000ng of RNA18s-1 cDNA to obtain reliable and verifiable results.
