Phytochemical study of leaves and fruits of Cavendishia nitida (Kunth) A. C. Sm and evaluation of its antioxidant activityrésumé
Cavendishia nitida (Kunth) A. C. Sm es un arbusto terrestre de 1 a 3 metros de altura, con tallo erecto y glabro. Se le conoce como “Uva camarona”, “uvo”, o “uva”. Es una planta endémica para Colombia, es posible encontrarla entre 1300 y 3000 msnm, en las Cordilleras Central y Oriental. En Colombia, los frutos son aprovechados como alimento por habitantes del altiplano cundiboyacense. Si bien Colombia cuenta con numerosas especies del género Cavendishia hay un vacío considerable de información con respecto a la química de estas. Por tanto, el objetivo del presente trabajo es contribuir al conocimiento químico de una especie de la familia Ericaceae endémica de Colombia (Cavendishia nítida (Kunth) A. C. Sm.), así como determinar su actividad antioxidante, con el fin de tener información que permita incentivar su potencial aprovechamiento. Para llevarlo a cabo, se colectaron hojas y frutos, posteriormente se prepararon extractos etanólico y metanólico de cada uno respectivamente. Luego, el extracto total de hojas fue fraccionado y subfraccionado mediante cromatografía en columna por gravedad. Por último, se realizó la elucidación estructural de los compuestos, usando cromatografía de gases acoplada a espectrometría de masas (GC-MS). Por otra parte, se determinó la actividad antioxidante del extracto total de hojas, la subfracción de Acetato de Etilo y la de Butanol junto con el extracto total de Frutos, mediante la capacidad captadora de radicales ABTS y DPPH. Finalmente de estos extractos y fracciones, se cuantificó el contenido de fenoles totales, flavonoides totales y antocianinas; mediante el método de Folin – Ciocalteu, Tricloruro de Aluminio e interpolación externa en HPLC – DAD, respectivamente. Se reporta la presencia de cuatro compuestos: Rimueno, Kaur-16-eno, α – amirina y β – amirina, por primera vez para esta especie. Adicionalmente, se encontró que de las fracciones obtenidas la que cuenta con mayor actividad antioxidante es la de Acetato de Etilo. Por otro lado, hay un mayor contenido de fenoles totales y flavonoides totales en el extracto total de hojas y las subfracciones analizadas. Finalmente, se encontró también que el extracto total de frutos es el que cuenta con un mayor contenido de antocianinas.
Abstrait
Cavendishia nitida (Kunth) A. C. Sm is a terrestrial shrub 1 to 3 meters high, with an erect and glabrous stem. It is known as "Uva camarona", "uvo", or "uva". It is an endemic plant for Colombia, it is possible to find it between 1300 and 3000 meters above sea level, in the Central and Eastern Cordilleras. In Colombia, the fruits are used as food by inhabitants of the Cundinamarca-Boyacá highlands. Although Colombia has numerous species of the genus Cavendishia there is a considerable gap in information regarding their chemistry. Therefore, the objective of the present work is to contribute to the chemical knowledge of a species of the family Ericaceae endemic to Colombia (Cavendishia nítida (Kunth) A. C. Sm.), as well as to determine its antioxidant activity, in order to have information that allows to stimulate its potential use. To do so, leaves and fruits were collected, and later ethanolic and methanolic extracts were prepared from each one respectively. Then, the total extract of leaves was fractionated and sub-fractionated by means of column chromatography by gravity. Finally, the structural elucidation of the compounds was performed, using gas chromatography coupled to mass spectrometry (GC-MS). On the other hand, the antioxidant activity of the total leaf extract, the subfraction of ethyl acetate and the subfraction of butanol together with the total fruit extract were determined by means of ABTS and DPPH radical capture capacity. Finally, from these extracts and fractions, the content of total phenols, total flavonoids and anthocyanins was quantified; by the method of Folin - Ciocalteu, Aluminum Trichloride and external interpolation in HPLC - DAD, respectively. The presence of four compounds is reported: Rimuene, Kaur-16-ene, α - amirin and β - amirin, for the first time in this species. Additionally, it was found that of the fractions obtained the one with the highest antioxidant activity is Ethyl Acetate. On the other hand, there is a higher content of total phenols and total flavonoids in the total extract of leaves and subfractions analyzed. Finally, it was also found that the total extract of fruits is the one with the highest content of anthocyanins.
